Nucleotide-sugars are the activated building blocks for the biosynthesis of carbohydrates, oligosaccharides, and glycan conjugates. Their metabolism is particularly well studied in yeast, fungal pathogens, and bacterial pathogens, such as Escherichia coli and Mycobacterium tuberculosis. These pathways are also studied in plants, but the enzymes involved are less well understood. Nucleotide-sugars act as glycosyl donors, forming components of the fungal and bacterial cell wall, serve as virulence factors, and modulate cell surface glycoprotein cellular activity. Furthermore, the alteration of oligosaccharide and glycan sequences have been studied in vaccines against bacterial or fungal infections, as well as, cancer. Considering the crucial roles nucleotide-sugars, oligosaccharides, and glycan conjugates are involved in, they are targets for drug development, and their analogs are potential inhibitors of these processes.
